原理

RNeasy技術(shù)將高效的異硫氰酸胍裂解技術(shù)與快速高純度的硅膠膜純化技術(shù)相結(jié)合，使總RNA分離變得簡(jiǎn)單化。該技術(shù)無(wú)需通過(guò)氯化銫（CsCl）梯度離心或氯化鋰/乙醇沉淀。RNeasy樣本制備技術(shù)已被*注冊(cè)，純化得到的核酸可用于任何分子檢測(cè)或其他下游應(yīng)用中，而無(wú)侵犯的風(fēng)險(xiǎn)。
RNeasy Plant Mini Kit提供QIAshredder分離柱（QIAshredder columns），用于勻質(zhì)化和通過(guò)離心法過(guò)濾高粘度的植物或真菌裂解物，同時(shí)采用RNeasy mini離心柱對(duì)RNA進(jìn)行純化。各種污染物，如：多聚糖等被洗掉，經(jīng)洗脫的RNA可直接用于各種下游應(yīng)用（見(jiàn)圖"Tissue Specificity of Histone H4 Expression"）。 RNeasy試劑盒提供高品質(zhì)RNA，幾乎無(wú)DNA殘留。對(duì)微量DNA非常敏感的RNA應(yīng)用，殘留的DNA可在RNeasy流程中使用RNase-Free DNase Set方便地通過(guò)DNA酶柱上消化去除。
操作流程
細(xì)菌樣品裂解后，在裂解液中加入乙醇來(lái)提供合適的結(jié)合條件，接著裂解液被上樣到RNeasy硅膠膜上。RNA與硅膠膜結(jié)合，其他各種污染物通過(guò)洗滌被有效去除。濃縮的純RNA被洗脫到無(wú)RNA酶的水中。
應(yīng)用
經(jīng)RNeasy技術(shù)純化得到的RNA，A260/A280比值可達(dá)1.9-2.1*，非常適合于各種下游應(yīng)用，包括：
