| 背景資料 | The protein encoded by this gene is the founding member of the protein tyrosine phosphatase (PTP) family, which was isolated and identified based on its enzymatic activity and amino acid sequence. PTPs catalyze the hydrolysis of the phosphate monoesters specifically on tyrosine residues. Members of the PTP family share a highly conserved catalytic motif, which is essential for the catalytic activity. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP has been shown to act as a negative regulator of insulin signaling by dephosphorylating the phosphotryosine residues of insulin receptor kinase. This PTP was also reported to dephosphorylate epidermal growth factor receptor kinase, as well as JAK2 and TYK2 kinases, which implicated the role of this PTP in cell growth control, and cell response to interferon stimulation. Two transcript variants encoding different isoforms have been found for this gene. |
多克隆抗體
抗原通常是由多個(gè)抗原決定簇組成的，由一種抗原決定簇刺激機(jī)體，由一個(gè)B淋巴細(xì)胞接受該抗原所產(chǎn)生的抗體稱之為單克隆抗體（Monclone antibody）。由多種抗原決定簇刺激機(jī)體，相應(yīng)地就產(chǎn)生各種各樣的單克隆抗體，這些單克隆抗體混雜在一起就是多克隆抗體（Polyclonal antibody），機(jī)體內(nèi)所產(chǎn)生的抗體就是多克隆抗體。
由于多克隆抗體通常包含抗某一抗原的不同表位的抗體，所以，在深固定的樣本中也會(huì)很好地發(fā)揮效應(yīng)，在石蠟包埋組織切片的染色中，多克隆抗體常選用。根據(jù)實(shí)驗(yàn)的不同需要，多克隆抗體應(yīng)用于相應(yīng)抗原的標(biāo)記。此外，在農(nóng)業(yè)生產(chǎn)中，多克隆抗體被用于農(nóng)藥殘留現(xiàn)場(chǎng)監(jiān)測(cè)；在臨床應(yīng)用中，多克隆抗體主要用于病原物的檢測(cè)、疾病的診斷及治療，如作為蛋白類免疫抑制劑用于移植反應(yīng)和自身免疫病的治療。
