Bovine serum albumin (BSA) is a slightly acidic ~66 kd protein. BSA is commonly used in biological research due to its low cost and inertness. Dilute enzymes can be stabilized by BSA (useful for restriction digests). BSA is a common blocking agent in western blotting and ELISA’s. Although, bovine serum albumin is exceptionally soluble (up to 50% w/v in buffered saline), it is typically used at a concentration of 0.1-1% w/v.

牛血清白蛋白,英文名稱為Bovine Serum Albumin,也稱Bovine albumin,或Cohn Fraction V,簡稱BSA。BSA有著其的實驗室應用,在實驗中作封閉劑,包括ELISA、WB(Western Blot)。作為載體蛋白,將其交聯(lián)于半抗原和其他弱抗原可以使它們在抗體中具有的原性。在限制性內(nèi)切消化反應中,BSA常常被用作一些的反應穩(wěn)定劑,并且能防止它粘附到管壁和槍頭上。BSA也于生物制的過程或者作為營養(yǎng)物用于和微生物培養(yǎng)。除此之外,還作為蛋白定量檢測的標準品使用。我們提供的BSA產(chǎn)品,使用牛血漿,利用熱休克法(Heat Shock)法制備而成。

蛋清中的主要蛋白質(zhì)包括卵清蛋白、卵轉(zhuǎn)鐵球蛋白、類卵沾蛋白、卵沾蛋白、溶菌、球蛋白G2、球蛋白G3等。卵清蛋白等電點4.5,其本質(zhì)是含磷糖蛋白。卵清蛋白是蛋清中的主要蛋白組分,其結構與性質(zhì)會影響蛋清蛋白在加工過程中的功能性質(zhì)。因此研究卵清蛋白在熱處理或美拉德反應后的變化對了解蛋清蛋白在加工過程中的結構與性質(zhì)的變化有意義。
卵清蛋白是一種蛋白質(zhì),占蛋清蛋白總量的54%-69%,卵清蛋白是的球蛋白,分子量為44.5kDa,屬含磷糖蛋白,含有四個自由巰基、385個氨基酸殘基 [1] 。這些氨基酸殘基相互纏繞折疊形成具有度二級結構的球型結構,大部分為α-螺旋和β-折疊。卵清蛋白有1個二硫鍵和4個巰基,當加熱時暴露出來,通過分子間相互作用使卵清蛋白膠體結構變得穩(wěn)定

該納米載體取材綠色、設計巧妙,可在部位依次針對表面過表達、微酸和谷胱甘肽(GSH)濃度等微實現(xiàn)三重響應性釋(圖2)。作者先通過定向解奶酪副產(chǎn)物-乳清蛋白獲得兩親性多肽,在水溶液中可自組裝形成30納米單分散膠束peptosome (PS),荷載疏水性抗姜黃素Cur,了姜黃素水溶液難溶的問題,并降低其對正常組織的副作用。隨后他們對其表面修飾功能肽段PePm,其中Pe可靶向表面的ErB2受體,且兼具的抗轉(zhuǎn)移效果。此外,Pm片段可以根據(jù)表面過表達金屬基質(zhì)蛋白MMP的特異性水解,通過交聯(lián)單個載體內(nèi)部自身巰基形成二硫鍵,使PS具有GSH濃度響應釋的功能。他們將所構建的載體PePm/PS/Cur經(jīng)尾靜脈注射到小鼠體內(nèi)后,經(jīng)EPR效應和Pe的靶向效應在部位度富集,載體表面Pe肽被MMP水解脫離,使Pe肽留在表面發(fā)揮抑和抗轉(zhuǎn)移效果。PS/Cur繼續(xù)通過“質(zhì)子海綿”效應從溶體中逃逸,在質(zhì)內(nèi)通過GSH響應逐漸解體,釋放出PS/Cur內(nèi)荷載的姜黃素,發(fā)揮抑制的作用。

豆球蛋白A(Con A),又名豆凝集素、豆素,是一種植物血凝素,具的有絲作用,有的淋巴轉(zhuǎn)化反應的作用,其淋巴轉(zhuǎn)化適濃度為40-100μg/ml,能沉淀糖原,凝集羊、馬、狗、兔、豬、大鼠、小鼠、豚鼠等及人紅。還能選擇性抑制性T(Ts),對調(diào)節(jié)機體反應具有作用。因此,通過使用Con A來活化態(tài)(或老年)時的Ts這一途徑,可望改觀一些自身性,甚或移植物排斥反應或惡性的防治。

Concanavalin A (con A) is a homotetrameric, 26 kDa lectin derived from the jack bean, Canavalia ensiformis. Concanavalin A binds to alpha-D glucose and alpha-D-mannose containing saccharides. Concanavalin A requires both Mn2+ and Ca2+ ions as a co-factor for binding. Since each con A has four binding sites, it can agglutinate cells such as red blood cells. Con A can also cause cell surface receptor aggregation which can be mitogenic for some lymphocytes.

Avidin is a tetrameric protein commonly isolated from egg whites. Avidin binds to biotin with strength approaching that of a covalent bond. Disruption of the avidin-biotin bond occurs only under extraordinary conditions, such as pH 1.5 with 6 M guanidium hydrocholoride. Fluorescent avidin can bind to biotinylated antibodies used in immunohistochemistry and immunocytochemistry. Avidin thiol can bind to gold surfaces and act as a glue for biotinylated proteins. Avidin agarose is useful for efficient removal of biotinylated proteins from a liquid phase.

Protein A has strong binding affinity towards immunoglobulins and it has been used for antibody identification, purification and many other biochemical research uses. Fluorescein protein A is a green fluorescent protein A conjugate that has excitation/emission wavelength at 495 nm/515 nm. 蛋白A(Protein A)是一種發(fā)現(xiàn)于金黃色葡萄球菌的壁表面蛋白,蛋白G(Protein G)是一種分離自G型或C型鏈球菌屬的表面蛋白,二者功能相似,主要通過與球蛋白(Ig)的Fc區(qū)相互作用,可結合大多數(shù)哺乳的IgG。然而兩者結合特異性上有所不同(詳細見附錄1,蛋白A,G對不同物種Ig的結合能力總表),如蛋白G對人IgG3,大鼠IgG2a,綿羊IgG1具有很的親和力,但蛋白A對這些Ig結合力很弱。蛋白A,蛋白G常共價偶聯(lián)在固體載體如瓊脂糖珠或丙烯酸樹脂小珠上,直接用以做沉淀(IP)或者共沉淀(Co-IP)來進行蛋白功能相關研究,或者直接裝在層析柱上來純化單克隆或者多克隆抗體。

Protein G is an immunoglobulin binding protein in Streptococci. Protein G binds to the Fc region of antibodies and thereby leaves the Fab region available to still bind antigen. Our recombinant version has been engineered to lack the albumin binding domains and has a 6x histidine tag at its N-terminus.

Green fluorescent protein (GFP) is a 27 kDa protein derived from Aequorea victoria. It absorbs light in the ultraviolet to blue range and emits green (peak emission 510 nm). GFP is a common reporter molecule for protein expression and visualization of intracellular processes. We produce this protein in E.coli.
