Kinetic-QCL™檢測(cè)是一種定量的，動(dòng)態(tài)檢測(cè)革蘭氏陰性細(xì)菌內(nèi)毒素。樣品與鱟試劑（LAL）混合，放在溫育板讀數(shù)器上，溶液變?yōu)辄S色指示測(cè)定完 成。Kinetic-QCL™顯色檢測(cè)特點(diǎn)用于微量注射藥物，疫苗，抗體和生物制劑檢測(cè)。相比于動(dòng)態(tài)濁度法和凝膠檢測(cè)法，Kinetic-QCL™ 不會(huì)干擾凝膠形成。檢測(cè)靈敏度范圍在0.005 EU/ml - 50.0 EU/ml，是所有鱟試劑（LAL）檢測(cè)方法中zui靈敏的。Kinetic-QCL™允許許多樣品在96高通量孔板中同時(shí)進(jìn)行檢測(cè)，節(jié)約時(shí)間和工作量。
Kinetic-QCL™（50-650U）包含8瓶LAL鱟試劑，2瓶標(biāo)準(zhǔn)內(nèi)毒素，3瓶LAL試劑水。每個(gè)試劑盒可以檢測(cè)192次。
§檢測(cè)原理：革蘭氏陰性細(xì)菌中內(nèi)毒素激活LAL中酶原，已激活的酶催化無色底物Ac-Ile-Glu-Ala-Arg-pNA裂解為肽段和硝基苯胺（pNA）。pNA可以在溫育板分光光度計(jì)405nm讀數(shù)，根據(jù)樣品反應(yīng)時(shí)間和標(biāo)準(zhǔn)物內(nèi)毒素反應(yīng)時(shí)間可以確定樣品中內(nèi)毒素濃度。
§結(jié)果處理：用微孔板讀數(shù)器測(cè)出每孔405nm吸光值（相應(yīng)空白孔作對(duì)照）。WinKQCLTM Software自動(dòng)生成標(biāo)準(zhǔn)物反應(yīng)時(shí)間和標(biāo)準(zhǔn)內(nèi)毒素濃度log/log線性相關(guān)。如果相關(guān)系數(shù)值≥0.980，則可以建立多項(xiàng)式模型制作標(biāo)準(zhǔn)曲線， 計(jì)算樣品內(nèi)毒素濃度。使用WinKQCLTM Software軟件可以線性回歸自動(dòng)計(jì)算出結(jié)果，具體計(jì)算過程參閱說明書。

LONZA Kinetic-QCL 192 Test Kit內(nèi)毒素檢測(cè)試劑盒 英文簡(jiǎn)述：
The Kinetic-QCL™ assay is a quantitative, kinetic assay for the detection of Gram negative bacterial endotoxin. A sample is mixed with the LAL/substrate reagent, placed in an incubating plate reader, and monitored over time （via WinKQCL™ Software） for the appearance of a yellow color.
It is the chromogenic nature of the Kinetic-QCL™ assay that makes it the most appropriate choice for testing small volume parenterals, vaccines, antibiotics and biologicals. The Kinetic-QCL™ assay is less impacted by inhibitory products which may interfere with the clotting mechanism in Turbidimetric and Gel Clot assays. With a sensitivity range of 0.005 EU/ml - 50.0 EU/ml, the Kinetic-QCL™ assay is the most sensitive of the LAL assays. Multiple samples can be processed at one time in this 96-well format resulting in time savings in a busy laboratory. An incubating plate reader is required to run this assay.

UltraCULTURE™ Serum-free Medium英文簡(jiǎn)述：
LONZA 12-725F UltraCULTURE™ Serum-free Medium is a complete, allpurpose medium designed for the c*tion of a wide variety of adherent and non-adherent mammalian cell types. UltraCULTURE™ Medium can be used to support fusion of cells during hybridoma formation, growth of monocyte, macrophage, epithelial, and fibroblastic cell lines, and generation of virus particles for vaccine production. The medium is supplemented with recombinant human insulin, bovine transferrin, and a purified mixture of bovine serum proteins. The total protein concentration of UltraCULTURE™ Medium is approximay 3 mg/ml. UltraCULTURE™ Medium can be supplemented with Cryoprotective Medium (Cat. No. 12-132A) to cryopreserve cells in a serum-free environment. UltraCULTURE™ Medium does not contain L-glutamine; please add 5 ml of 200 mM L-glutamine solution (Cat. Nos. 17-605 or 17-905) prior to use.
