| I. Background | |
| 白細(xì)胞介素 22（IL-22）是 IL-10 細(xì)胞因子家族的成員，是一種新興的 CD4+Th 細(xì)胞因 子，在抗微生物防御、體內(nèi)平衡和組織修復(fù)中發(fā)揮重要作用。該家族還包括其他細(xì)胞因子 如 IL-10、IL-19、IL-20、IL-24、IL-26 和 III 型干擾素（IFNλ）。IL-22 是一種主要由淋巴 細(xì)胞分泌的細(xì)胞因子；包括先天性和適應(yīng)性免疫系統(tǒng)，例如 3 型固有淋巴細(xì)胞（ILC3）、 αβT 細(xì)胞、γδT 細(xì)胞和 NKT 細(xì)胞。在沒(méi)有明顯感染或炎癥的情況下，即處于穩(wěn)定狀態(tài)，IL- 22 的主要來(lái)源是 ILC3，它大量存在于大腸和小腸的黏膜中。在 αβT 細(xì)胞中，產(chǎn)生 IL-22 的 主要是 Th1、Th17、Th22 細(xì)胞；大約 33% 的產(chǎn)生 IL-22 的 CD4+ T 細(xì)胞 Th1 細(xì)胞，50% 是 Th22 細(xì)胞，15% 是 Th17 細(xì)胞。中性粒細(xì)胞也可產(chǎn)生 IL-22 。 功能性 IL-22R 是由 IL-22R1 和 IL-10R2 組成的異二聚體，IL-10R2 與 IL-10 家族的各 種細(xì)胞因子受體復(fù)合物共有。IL-10R2 普遍表達(dá)，而 IL-22R1 僅在如支氣管、肝臟、胰腺 和腸道和基質(zhì)細(xì)胞的表皮細(xì)胞中表達(dá)。在肝臟中，肝細(xì)胞同時(shí)表達(dá) IL-10R2 和 IL-22R1； 許多疾病，如牛皮癬、移植物抗宿主病、肝臟和胰腺損傷、潰瘍性結(jié)腸炎和腫瘤與 IL-22 密切相關(guān)。 在 IL-22 與 IL-22 受體結(jié)合后，Jak1/Tyk2 激酶被激活，導(dǎo)致這些受體和轉(zhuǎn)錄激活因子 STAT1、STAT3、STAT5 蛋白的磷酸化。STAT3 誘導(dǎo)涉及多種信號(hào)通路的許多基因的表達(dá)， 包括與細(xì)胞凋亡、細(xì)胞周期等有關(guān)的通路。除了 STAT 信號(hào)傳導(dǎo)外，IL-22-IL-22R1-IL- 10R2 復(fù)合物還導(dǎo)致 MAPK 和 p38 通路的激活。 | |
