| I. Background | |
| 生長分化因子 15（GDF15），也稱為巨噬細(xì)胞抑制性細(xì)胞因子 1（MIC-1），屬于 TG F-β超家族蛋白。GDF15 是一種內(nèi)分泌激素，主要參與細(xì)胞生長、分化和組織修復(fù)。GDF1 5 作為一種炎癥標(biāo)志物，在腫瘤、缺血性疾病、代謝紊亂和神經(jīng)退行性過程的發(fā)病機(jī)制中 發(fā)揮作用。 GFRAL（GDNF-family receptor α-like），也被稱為 GRAL，屬于膠質(zhì)細(xì)胞源性神經(jīng)營 養(yǎng)因子家族α樣受體。GFRAL 是一種僅在后腦中表達(dá)的跨膜蛋白，需要輔助蛋白 RET 的幫 助才能進(jìn)行信號傳導(dǎo)。當(dāng) GDF15 與 GFRAL 結(jié)合時(shí)，RET 將自磷酸化并導(dǎo)致下游信號通路 （如 PI3K-AKT 和 PLC-PKC）的激活，從而調(diào)節(jié)許多生理過程，包括影響食物攝入和導(dǎo)致 體重減輕。抑制 GDF15-GFRAL 信號通路則能夠增強(qiáng)免疫系統(tǒng)對實(shí)體瘤的殺傷，并能夠治 療或預(yù)防癌癥惡病質(zhì)。 | |
