背景 Prolactin is synthesis starts in first few weeks of gestation in pituitary lactotropes and serum levels decline shortly after birth. Prolactin levels rise significantly during pregnancy. Other factors that regulate Prolactin levels include sleep, stress, hypoglycemia and exercise. Prolactin is structurally related to growth hormone and placental lactogen. The genes for these hormone share common structural homology, prolactin is coded by a single gene located on human chromosome 6. The prolactin s a single polypeptide of 199 amino acids with a molecular mass of 23?kDa and has 2 disulfide bond, one more than GH. Majority of Prolactin is in circulation in monomeric form (23?kDa). Unlike GH, the prolactin is glycosylated at asparagine. Some dimeric, polymeric and degraded form of prolactin is also detectable in circulation, but their physiological significance is not known. Prolactin binds to two prolactin receptors, with downstream signal transduction mediated by tyrosine kinase cascade. Physiological effects include breast development, and effects on hypothalamus-pituitary-gonadal axis. Prolactin receptor are also present in T lymphocytes, and prolactin has been shown to modulate immune responses (1). Prolactin secreting tumors are common cause of hyperprolactinemia, in women the symptoms relate to galactorrhea, amenorrhea and infertility. In men the symptoms include galactorrhea, infertility and impotance.
