The Cellartis Hepatocyte Differentiation Kit用于誘導(dǎo)內(nèi)胚層細胞（definitive endoderm (DE)）獲取肝細胞。通過使用本產(chǎn)品獲取的肝細胞表現(xiàn)出符合預(yù)期的代謝酶表達，從而可以用于藥物代謝研究和毒理學篩選。本產(chǎn)品包含足夠的維持培養(yǎng)基用于正常為期11天的實驗。
The Cellartis Hepatocyte Differentiation Kit可以對廣泛來源的DE細胞進行定向分化：客戶自己的DE細胞，或是Cellartis提供的標準化DE細胞，或是客戶利用Cellartis Definitive Endoderm Differentiation Kit獲得的DE細胞。據(jù)估計，現(xiàn)在已經(jīng)有28種發(fā)源于人類干細胞的DE細胞被成功測試可以使用本產(chǎn)品。特別地，如果客戶是用于大規(guī)模、要求持續(xù)穩(wěn)定獲得肝細胞的相關(guān)用途，Cellartis推薦Definitive Endoderm Cells (from ChiPSC18)，用作起始DE細胞材料，或是用作陽性對照品。如果客戶要求的DE是來自于自有的樣本材料（patient-specific, or disease-specific），建議使用Cellartis iPS Cell to Hepatocyte Differentiation System (Cat. The Cellartis Hepatocyte Differentiation Kit generates highly homogeneous hepatocyte cultures starting from definitive endoderm (DE) cells. The resulting hepatocytes show the expected expression of relevant metabolic enzymes, making them suitable for drug metabolism studies and toxicity screening. The Cellartis Hepatocyte Differentiation Kit contains sufficient maintenance medium to perform experiments in an 11-day time window.
There are several options for source materials to generate functional hepatocytes: your own DE cells, our DE cells, or DE cells you generate using the Cellartis Definitive Endoderm Differentiation Kit. This kit has been successfully tested with DE cells derived from 28 different human pluripotent cell lines. Our cells, Cellartis Definitive Endoderm Cells (from ChiPSC18), are suitable as the starting material or as a positive control. If you desire a customized (standard, patient-specific, or disease-specific) genetic background for your DE cells, use the complete Cellartis iPS Cell to Hepatocyte Differentiation System (Cat. No. Y30055).

Definitive endoderm (DE) cells were differentiated into hepatocytes using the Cellartis Hepatocyte Differentiation Kit. The first field of view, labeled “Definitive Endoderm Day 7,” shows DE cells seven days after differentiation from human pluripotent cells. These DE cells became the starting material for the differentiation kit. The remaining progression from DE cells to hepatocytes shows the expected morphology over a 14-day time period. The morphological features present in the last field of view, labeled “Hepatocyte Day 21+,” continue stably past Day 21.
