腫瘤細(xì)胞實(shí)驗(yàn)原理
閱讀:161 發(fā)布時(shí)間:2017-4-17腫瘤細(xì)胞增殖是生物體的重要生命特征,細(xì)胞以分裂的方式進(jìn)行增殖。單細(xì)胞生物,以細(xì)胞分裂的方式產(chǎn)生新的個(gè)體。多細(xì)胞生物,以細(xì)胞分裂的方式產(chǎn)生新的細(xì)胞,用來(lái)補(bǔ)充體內(nèi)衰老和死亡的細(xì)胞;同時(shí),多細(xì)胞生物可以由一個(gè)受精卵,經(jīng)過(guò)細(xì)胞的分裂和分化,zui終發(fā)育成一個(gè)新的多細(xì)胞個(gè)體。必須強(qiáng)調(diào)指出,通過(guò)細(xì)胞分裂,可以將復(fù)制的遺傳物質(zhì),平均地分配到兩個(gè)子細(xì)胞中去??梢?jiàn),腫瘤細(xì)胞增殖是生物體生長(zhǎng)、發(fā)育、繁殖和遺傳的基礎(chǔ)。真核細(xì)胞的分裂方式有三種,即有絲分裂、無(wú)絲分裂和減數(shù)分裂。
活細(xì)胞線(xiàn)粒體中的琥珀酸脫氫酶能使外源性MTT還原為水不溶性的藍(lán)紫色結(jié)晶甲瓚(Formazan)并沉積在細(xì)胞中,而死細(xì)胞無(wú)此功能。然后采用二甲基亞砜(DMSO)溶解細(xì)胞中的甲瓚,用酶聯(lián)免疫檢測(cè)儀在570 nm波長(zhǎng)處測(cè)定其光吸收值,反映活細(xì)胞數(shù)量。在一定細(xì)胞數(shù)范圍內(nèi),MTT結(jié)晶形成的量與腫瘤細(xì)胞數(shù)成正比。
