點(diǎn)擊化學(xué)又譯為“鏈接化學(xué)”、“速配接合組合式化學(xué)”,是由化學(xué)家巴里·夏普萊斯(K B Sharpless)在2001年引入的一個(gè)合成概念,主旨是通過小單元的拼接,來快速可靠地完各種分子的化學(xué)合成。它尤其強(qiáng)調(diào)開辟以碳-雜原子鍵(C-X-C)合成為基礎(chǔ)的組合化學(xué)新方法,并借助這些反應(yīng)(點(diǎn)擊反應(yīng))來簡單高效地獲得分子多樣性。點(diǎn)擊化學(xué)的代表反應(yīng)為銅催化的疊氮-炔基Husigen環(huán)加成反應(yīng)(Copper-Catalyzed Azide–Alkyne Cycloaddition)。點(diǎn)擊化學(xué)的概念對(duì)化學(xué)合成領(lǐng)域有很大的貢獻(xiàn),在藥物開發(fā)和生物醫(yī)用材料等的諸多領(lǐng)域中。
點(diǎn)擊化學(xué)以其條件溫和、選擇性強(qiáng)等優(yōu)點(diǎn),在生物結(jié)合、生物標(biāo)記和材料科學(xué)等領(lǐng)域得到了廣泛的應(yīng)用,目前主要有以下幾個(gè)方面的應(yīng)用:
