TMB Plus Liquid 1-Component Substrate is a ready-to-use soluble substrate for kinetic and endpoint assays of horseradish peroxidase (HRP) in microwell procedures, such as ELISA.  Reaction of the substrate with HRP generates a free-radical cation that exhibits blue color with an absorption maximum at 653 nm.  Acidification of the reaction by addition of sulfuric acid yields a yellow-colored diimine terminal oxidation product, which absorbs light at 450 nm.  TMB Plus Liquid 1-Component Substrate is optimized with TMB and hydrogen peroxide concentrations that allow a linear response to typical HRP concentrations utilized in immunologic assays.
