【蛋白交聯(lián)劑的分類】:
1:同源交聯(lián)劑:
具有兩個(gè)相同活化反應(yīng)基團(tuán)。應(yīng)用:一步反應(yīng),分子內(nèi)交聯(lián),多聚反應(yīng),穩(wěn)定四級(jí)結(jié)構(gòu)
2:異源交聯(lián)劑:
具有兩個(gè)不同活化反應(yīng)基團(tuán)。應(yīng)用:兩步反應(yīng)交聯(lián),可減少聚集和自身交聯(lián)。反應(yīng)原則:先反應(yīng)不穩(wěn)定活性基團(tuán)(與*種蛋白質(zhì)),然后去除掉未反應(yīng)的交聯(lián)劑,在加入種待交聯(lián)蛋白質(zhì),短臂用于分子間交聯(lián),長(zhǎng)臂用于分子內(nèi)交聯(lián)反應(yīng):SH……NH2, NH2…… COOH
3:光反應(yīng)交聯(lián)劑:
反應(yīng)原則:先交聯(lián)不穩(wěn)定的熱反應(yīng)活性基團(tuán)(與*種蛋白質(zhì)),然后UV交聯(lián),將*種蛋白質(zhì)交聯(lián)到種蛋白質(zhì)的N-H,C-H上。特點(diǎn):交聯(lián)非特異性,產(chǎn)率低。
【交聯(lián)劑的使用方法】:
圖示為同源交聯(lián)劑的使用方法:先將待交聯(lián)的蛋白質(zhì)A與交聯(lián)劑在一定條件下反應(yīng),得到的蛋白質(zhì)A-交聯(lián)劑中間體,然后移除多余的交聯(lián)劑,再加入欲交聯(lián)的蛋白質(zhì)B。產(chǎn)物中可能會(huì)形成多聚體,這一副產(chǎn)物通過(guò)調(diào)節(jié)反應(yīng)條件來(lái)控制。分成兩部反應(yīng)的目的在于減少分子內(nèi)交聯(lián)以及形成蛋白質(zhì)多聚物。另外,選擇異源交聯(lián)劑也可以減少多聚物的產(chǎn)生。
