DSS誘導(dǎo)斑馬魚潰瘍性結(jié)腸炎模型的建立

斑馬魚作為腸道損傷模型的應(yīng)用已得到充分證實(shí),葡聚糖硫酸鈉(Dextran Sulfate Sodium Salt,DSS)誘導(dǎo)的斑馬魚IBD模型模擬了哺乳動(dòng)物IBD表型的不同特征,包括腸道中性粒細(xì)胞炎癥、過多粘液生成、腸淋巴管生成增加以及前炎性細(xì)胞因子上調(diào)。

實(shí)驗(yàn)材料


DSS(Yeasen 60316ES,MW:36000~50000)

 


實(shí)驗(yàn)步驟


                                       1. 將斑馬魚胚胎培養(yǎng)在含甲基藍(lán)的E3胚胎培養(yǎng)基中,28.5 ℃,培養(yǎng)至1 dpf;

                                       2. 1dpf 后使用不含亞甲基藍(lán)的E3胚胎培養(yǎng)基,培養(yǎng)至3 dpf;

                                       3. E3培養(yǎng)基配制濃度為10% DSS的儲(chǔ)存液;

                                       4. 用培養(yǎng)基稀釋DSS至最大非致死劑量(DSS濃度參考:0.5%);

                                       5. 0.5%的DSS處理斑馬魚,從3 dpf處理到6 dpf,觀察指標(biāo)[1]。

 


結(jié)果分析


                                       造模成功指標(biāo):中性粒細(xì)胞數(shù)量增多、酸性粘液蛋白增多、炎性因子表達(dá)量上升。

                                       1.嗜中性粒細(xì)胞數(shù)量:中性粒細(xì)胞數(shù)量增多,并且向腸和上皮中移動(dòng)

                                       2.酸性粘液蛋白:石蠟病理切片結(jié)合AB-PAS染色法驗(yàn)證,DSS浸泡后斑馬魚腸道中酸性粘液蛋白增多

                                       3.炎性因子表達(dá)量DSS處理后斑馬魚腸道中炎性因子表達(dá)量上升[1,2]

DSS處理組相較于對(duì)照組,酸性粘蛋白染色和粘液分泌增加,斑馬魚外周血中的嗜中性粒細(xì)胞數(shù)量增多并且向腸和上皮中移動(dòng),炎性因子表達(dá)量顯著上調(diào),腸道出現(xiàn)炎癥反應(yīng)。
