DSS誘導(dǎo)果蠅潰瘍性結(jié)腸炎模型的建立

葡聚糖硫酸鈉(Dextran Sulfate Sodium,DSS)是葡聚糖的聚陰離子衍生物,喂食果蠅會(huì)破壞腸上皮中腸道表皮細(xì)胞之間的連接從而造成腸道損傷,引起多條通路應(yīng)答調(diào)控干細(xì)胞的自我更新及分化,補(bǔ)充受損細(xì)胞,維持腸道穩(wěn)態(tài)。

實(shí)驗(yàn)動(dòng)物


5-10日齡果蠅,雌性 

 


實(shí)驗(yàn)材料


DSS(yeasen DSS 60316ES,MW36000-50000) 

 


實(shí)驗(yàn)步驟


1. 用5%的蔗糖溶液配制喂食培養(yǎng)基;
2. 對(duì)照組:5%蔗糖培養(yǎng)基(SC),實(shí)驗(yàn)組:3% DSS(培養(yǎng)基配制);
3. 預(yù)先用500 μL的喂食培養(yǎng)基將2.5cm×3.75cm的層析紙潤(rùn)濕,然后放入空的培養(yǎng)瓶中;
4. 果蠅放入含有層析紙的瓶中,29℃培養(yǎng)三天,期間每天將存活的果蠅移入含有新鮮培養(yǎng)基的空瓶中;
5. 培養(yǎng)三天后,切開中腸,用GFP(綠色)和PH3(紅色)抗體和DRAQ5(藍(lán)色)進(jìn)行免疫染色。

2.  DSS誘導(dǎo)果蠅中ISC前體細(xì)胞增殖[1]

 通過免疫染色中不同的標(biāo)記方法,量化了腸道干細(xì)胞(ISC),ISC前體細(xì)胞發(fā)生增值,表明DSS誘導(dǎo)果蠅模型腸道發(fā)生損傷。
