淋巴細胞分離液
LYMPHOCYTE SEPARATION MEDIUM
LSM的優(yōu)勢：
傳統(tǒng)的組合是Ficoll+泛影酸葡甲胺（甲泛影鈉）。Yeasen公司的LSM主要成分是Ficoll （聚蔗糖）和sodium diatrizoate （泛影酸鈉，密度1.200±0.002g/ml），每100ml包含6.2g聚蔗糖和9.4g泛影酸鈉。目前為止，這個組合是淋巴細胞分離液成分使用zui廣泛、分離效果的組合。
與LSM競爭zui大的產(chǎn)品主要是：GE公司的Ficoll-Paque PREMIUM，其特點如下：

使用說明（用于去纖維蛋白或抗凝血劑處理過的人體血液；對于其他物種或者其他組織的血液，需要對此程序做出一定的改進）
1. 輕輕顛倒瓶子使LSM充分混合
2. 無菌轉(zhuǎn)移3 mlLSM到15 ml離心管中。
3. 混合2 ml除纖維蛋白血液或肝素處理血液 和2 ml 生理鹽水
4. 仔細的將稀釋血液加入3 mlLSM（室溫）于15ml離心管中，在血液和LSM中形成一個明顯的分層。不要將稀釋血液混合入LSM中。
5. 室溫下400g離心15-30分鐘，離心可以沉淀紅細胞和多核白細胞同時可以再LSM上形成一層單核淋巴細胞，如下圖所示。
6. 吸出淋巴細胞上方2-3mm的血漿。
7. 吸取淋巴細胞層以及它下面一半的LSM和轉(zhuǎn)移到另外的一個離心管。加入等體積的平衡鹽緩沖液至淋巴細胞離心管中，室溫離心10分鐘，離心速度設(shè)定在既不損傷細胞又能沉淀細胞即刻，例如160 - 260 x g（去除LSM和降低血小板的百分比）。
8. 用平衡鹽緩沖液清洗細胞，用適當?shù)呐囵B(yǎng)基重懸細胞。
