這可能與二種抑制 劑和DP-TP載體的結合部位不同有關。蒼術苷只能與DP-TP載 體的胞液側結合而米酵菌酸可與DP-TP載體的胞液及基質(zhì)二側結 合。線粒體作用⑴若將純化的正常的線粒體與純化的細胞核在一起保溫，并不導致 細胞核的變化。但若將誘導生成PT孔道的線粒體與純化的細胞核一 同保溫，細胞核即開始凋亡變化。⑵細胞死亡調(diào)節(jié)蛋白不論是抑制 死亡的bcl-家族還是促進細胞死亡的Bx家族均以線粒體作為靶 細胞器。bcl-蛋白的C端的疏水肽段能插入線粒體外膜。事實上 相當量的bcl-位于線粒體內(nèi)外膜的接觸位點。⑶高表達bcl-能 防止ΔΨm的耗散，從而導致對蒼術苷、原卟啉IX與mClCCP的不敏 感與IF釋放的抑制；反之，高表達Bx則導致ΔΨm的耗散。綜上所述，細胞凋亡與線粒體的結構與功能有著密切的關系。如果 線粒體有大量PT孔道形成，細胞TP濃度很快下降，則在致凋亡的 蛋白酶被活化前細胞就壞死了。而如果PT孔道的誘導生成是一種比 較緩和與持續(xù)的狀態(tài)，在細胞TP濃度下降前專一的蛋白酶被激活 ；而另一方面ΔΨm的耗散產(chǎn)生的超氧陰離子則導致細胞死亡。細 胞凋亡是一把劍。
This may be related to the different binding sites of the two inhibitors and the DP-TP vector. Atractylenosides bind only to the cytosolic side of the DP-TP carrier and rice yeasts bind to the cytosolic and matrix sides of the DP-TP carrier. Mitochondrial action (1) Incubation of purified normal mitochondria with purified nuclei does not result in changes in the nucleus. However, if the mitochondria that induce the formation of PT pores are incubated with purified nuclei, the nuclei start to undergo apoptosis. (2) The cell death regulatory protein, either the bcl-family that inhibits death or the Bx family that promotes cell death, uses mitochondria as target organelles. The C-terminal hydrophobic peptide of the bcl-protein can be inserted into the mitochondrial outer membrane. In fact, a considerable amount of bcl- is located in the mitochondrial outer membrane contact sites. (3) High expression of bcl- can prevent the dissipation of ΔΨm, resulting in insensitivity and inhibition of IF release of atractyloside, protoporphyrin IX and mClCCP; conversely, high expression of Bx leads to dissipation of ΔΨm. In summary, apoptosis has a close relationship with the structure and function of mitochondria. If mitochondria have a large number of PT pores formed, the concentration of TP in cells rapidly decreases, and the cells become necrotic before the proapoptotic proteases are activated. However, if the induction of PT channels is a more gradual and sustained state, specific proteases are activated before the concentration of TP in the cells declines; on the other hand, the superoxide anion produced by the dissipation of ΔΨm leads to cell death. Cell apoptosis is a sword.
