【檢測(cè)原理】

寨卡病毒IgG/IgM檢測(cè)試劑盒（膠體金法）采用膠體金免疫層析技術(shù)，在玻璃纖維紙上預(yù)包被金標(biāo)記摘卡重組抗原，在硝酸纖維膜上檢測(cè)線和對(duì)照線處分別包被抗人IgGΥ鏈單克隆抗體、抗人IgMμ鏈單克隆抗體和兔抗摘卡抗體。檢測(cè)陽(yáng)性樣本時(shí)，樣本血清（漿）中寨卡抗體（IgG或IgM）與膠體金標(biāo)記寨卡重組抗原(Au-Dengue-Ag)結(jié)合形成復(fù)合物，由于層析作用復(fù)合物沿紙條向前移動(dòng)，經(jīng)過檢測(cè)線時(shí)與預(yù)包被的抗人IgGΥ鏈單克隆抗體或抗人IgMμ鏈單克隆抗體形成“Au-Zika-Ag-抗Zika抗體-抗人IgGΥ（或抗人IgMμ）-固相材料”夾心物而凝聚顯色，游離金標(biāo)抗原則在對(duì)照線處與兔抗寨卡抗體結(jié)合而富集顯色。陰性標(biāo)本則僅在對(duì)照線處顯色。檢查中把經(jīng)過適當(dāng)稀釋的血清（漿）標(biāo)本加到檢測(cè)卡的加樣處，20分鐘內(nèi)觀察結(jié)果即可。

【使用方法】

1．用加樣器吸取血清或者血漿(漿)2ul，加在加樣孔的上端（S1）處；

2．在加樣孔（S）處滴加100ul（2滴）樣品稀釋液；

3．20分鐘內(nèi)觀察并記錄結(jié)果。

【對(duì)試驗(yàn)結(jié)果的解釋】

陽(yáng)性：1、IgG 陽(yáng)性：在IgG檢測(cè)線處和對(duì)照線位置出現(xiàn)兩條紫紅線。

2、IgM 陽(yáng)性：在IgM檢測(cè)線處和對(duì)照線位置出現(xiàn)兩條紫紅線。

3、IgG、IgM 同時(shí)陽(yáng)性：在IgG檢測(cè)線處、IgM檢測(cè)線處和對(duì)照線位置同時(shí)出現(xiàn)三條紫紅線。

陰性：只在對(duì)照線位置出現(xiàn)一條紫紅線。

無(wú)效：在對(duì)照線位置沒有出現(xiàn)紫紅線。

【注意事項(xiàng)】

1. 檢測(cè)必須符合傳染病實(shí)驗(yàn)室管理規(guī)范和生物安全守則的規(guī)定。所有樣品和廢棄物都應(yīng)按傳染物處理；

2．操作應(yīng)嚴(yán)格按說明書進(jìn)行；

3．實(shí)驗(yàn)環(huán)境應(yīng)保持一定濕度，避風(fēng)，避免在過高溫度下進(jìn)行實(shí)驗(yàn)（*在10～37℃）；

4．檢測(cè)試劑從鋁箔袋中取出后，應(yīng)盡快進(jìn)行實(shí)驗(yàn)，避免放置于空氣中過長(zhǎng)時(shí)間，試劑吸潮后將失效；

5．試劑盒可在室溫下保存，謹(jǐn)防受潮，低溫下保存的試劑盒應(yīng)平衡至室溫方可打開鋁箔袋進(jìn)行檢測(cè)操作；

6．為了防止弱陽(yáng)性標(biāo)本的漏檢，應(yīng)在加樣后20 分鐘判讀結(jié)果，在檢測(cè)線處出現(xiàn)紫紅線，即可判斷為陽(yáng)性結(jié)果，超過20 分鐘后結(jié)果只作為參考。
